The Molecular Weight Law of Exfoliation
The speed and depth at which an acid penetrates the skin is inversely proportional to its molecular size. A tiny acid molecule slips rapidly between corneocytes, releasing hydrogen ions deep in viable epidermis and provoking inflammatory mast cell degranulation.
In contrast, high-molecular-weight acids diffuse slowly and uniformly, exfoliating solely the outermost stratum corneum layers without triggering erythema.
1. Gluconolactone (PHA, 178 Daltons) — The Gentlest Exfoliant
Gluconolactone is a polyhydroxy acid (PHA) with a substantial molecular weight of 178.14 g/mol and multiple water-attracting hydroxyl groups.
Because of its bulky molecular ring, it cannot penetrate rapidly past superficial corneocytes. It provides gentle, uniform surface desquamation while functioning as a humectant and antioxidant, causing virtually zero stinging even in rosacea-prone skin.
2. Mandelic Acid (AHA, 152 Daltons) — The Melanin-Safe AHA
Mandelic acid is an alpha-hydroxy acid derived from bitter almonds. With a molecular weight of 152.15 Daltons, it is exactly double the size of glycolic acid.
This larger structure ensures slow, metered epidermal transit. Crucially, its gentle non-inflammatory profile makes it the safest AHA for Fitzpatrick skin types IV–VI, minimizing the risk of secondary post-inflammatory hyperpigmentation.
3. Lactic Acid (AHA, 90 Daltons) — Hydrating Surface Smoother
Lactic acid (90.08 Daltons) is an intermediate-sized AHA that naturally occurs in human skin as part of the natural moisturizing factor (NMF).
At concentrations of 5% to 10%, it cleaves calcium-dependent desmosomes while stimulating endogenous ceramide synthesis in keratinocytes, providing smooth texture with built-in hydration.
4. Salicylic Acid (BHA, 138 Daltons) — The Pore-Penetrating Lipophile
Unlike water-soluble AHAs, salicylic acid is a beta-hydroxy acid with an ortho-hydroxy aromatic structure that makes it completely lipid-soluble (lipophilic).
It diffuses directly through sebum-filled sebaceous follicles, solubilizing the keratinized plugs that cause blackheads and whiteheads. It also possesses inherent anti-inflammatory salicylate properties.
5. Glycolic Acid (AHA, 76 Daltons) — The High-Potency Powerhouse
Glycolic acid is the smallest alpha-hydroxy acid in existence (76.05 Daltons). Its minuscule two-carbon backbone allows it to rapidly penetrate deep into the epidermis and upper papillary dermis.
It delivers dramatic improvements in cellular turnover and collagen synthesis, but carries the highest potential for chemical burns, acid erythema, and barrier collapse if overused or combined with retinoids.
