Biological Mechanism of Irritation

Isothiazolinones and formaldehyde releasers act as potent low-molecular-weight electrophilic haptens. Methylisothiazolinone undergoes covalent addition with nucleophilic thiol (-SH) groups on epidermal cysteine residues, forming stable immunogenic neo-antigens. Formaldehyde releasers slowly hydrolyze in water-based formulations, liberating free gaseous formaldehyde (HCHO) at 100–300 ppm. This reactive gas cross-links lysine amino groups in skin proteins, activating Langerhans cells and driving memory T-lymphocyte proliferation (Type IV delayed hypersensitivity).
