Propylene glycol (1,2-propanediol) possesses amphiphilic solvent properties that transiently fluidize the stratum corneum lipid bilayers to accelerate transdermal active penetration. However, at concentrations exceeding 10–20%, its intense hygroscopic capacity draws water osmotically out of viable keratinocytes faster than microcapillary flow can replenish it. This creates non-immunological chemical irritant contact dermatitis, disruption of membrane potentials, and direct activation of sensory nerve endings.
